原理就是利用蛋白质在氯仿中变性来分离DNA和蛋白，并靠DNA在高浓度醇溶液中溶解性下降来纯化DNA。

DNA存在于白细胞中 所以红细胞和蛋白质是我们要去除的 首先是去除红细胞先是利用水的张力红细胞就可以去掉 但为了去除的彻底还需要加入盐溶液进一步清洗 之后就是要加入缓冲液和蛋白酶来去除蛋白质了 为了使蛋白质能够彻底去除 需要65°C水浴最好1个小时 这样得到的DNA纯度会提高 然后就是加入异丙醇或者是无水乙醇来沉淀DNA了 异丙醇沉淀小分子DNA比较合适 而无水乙醇是沉淀大分子DNA的
